Holmes was born in Toledo, Ohio. The youngest of five children born to housewife and philanthropist Kathleen A. Stothers and Martin Joseph Holmes Sr., a lawyer playing basketball at Marquette University under her coach, Al McGuire. He has three sisters and a brother. Holmes was baptized as a Roman Catholic and attended the King Christ Church in Toledo.
